I have a fresh install of 10.04 on an EeePC 1005HA and have the problem with no microphone sound in skype. Also in sound preferences the volume appears to be set at zero and cannot be adjusted. Sound recorder works OK however. Installed pulseaudio volume control and found that moving the slider bar for volume control restored microphone on skype and in sound preferences.
