I managed to fix the problem by editing 
/etc/cups/ppd/Officejet_Pro_8500_A910.ppd
Changing:
*UIConstraints: *Duplex *OptionDuplex False
*UIConstraints: *OptionDuplex False *Duplex
on line 36 to
*UIConstraints: *Duplex *OptionDuplex True
*UIConstraints: *OptionDuplex True *Duplex
gave me the two-sided options I was missing in the dialog.

With 
lpr -o sides=two-sided-long-edge document.pdf
it works.
My guess is that it would work if only the option was available in the printing 
dialog.

Thank you for your response Till. I tried different programs but it is
not possible to set two-sided in the printing dialog, so I believe that
is where the problem is. Maybe I can test with this option set from the
command line.

I am also affected by this bug using version hplip 3.11.10 and
Officejet_Pro_8500_A910. It is not possible to choose Two Sided in the
printer dialog box. Under Installable Options in hp-toolbox the option
Duplexer Installed is set to On but greyed out. Changing Double-Sided
Printing to Short Edge or Long Edge doesn't help. Also, the Page Size
A4 AutoDuplex is not available. Can't really see why you would need
the cups error_log but I will attach it as you request.

I got it working using the OSS Driver instead of the ALSA Driver in
winecfg. Wouldn't call it a bug fix, but at least I can use Spotify as
normal again.

I think increasing the nice-level in /etc/pulse/daemon.conf made the
problem go away. I set it to 3 instead of -11.
