Dear Colleagues,

I would like to draw your attention to the upcoming EMBO Course on 
“Membrane protein expression, purification, and characterization 3 ( 
mPEPC3) ”, which will take place from

9th until 16th September 2024 at the *EMBL Hamburg Unit*.

This EMBO Practical Course trains PhD students and early career postdocs 
with the latest developments in membrane protein production, 
characterization and structure determination methods. Modern protein 
expression techniques in various hosts will be presented and students 
will get acquainted with state-of-the-art approaches for studying 
structural and dynamic aspects of membrane proteins. An attitude to 
combine protein purification and characterization will be taught to 
impress the necessity on the students that proteins have to be 
conditioned with care to achieve structural information. The presence of 
a crystallization and cryo-EM facility as well as beamlines for SAXS and 
macromolecular crystallography at the venue will be employed to perform 
integrated structural biology experiments. The course has an emphasis on 
practicals in small groups, and we will ensure a close interaction 
between students and tutors.


The aim is to inspire the students to apply the newly acquired knowledge 
and to maintain interactions among themselves and the tutors afterwards.

I would like to share our new software for data analysis of Molecular 
Biophysics experiments The platform enables users to upload and analyse 
raw data from several experimental techniques: steady-state fluorescence 
spectroscopy, microscale thermophoresis, and differential scanning 
fluorimetry experiments. It consists of three modules. FoldAffinity 
quantifies how an interaction between two molecules can further 
stabilise them. MoltenProt helps to estimate how durable a protein is in 
different conditions. ThermoAffinity identifies how strongly two 
molecules, e.g. a protein and a drug, bind to each other. It is freely

EMBL beamlines ( 2x MX and 1 SAXS). An extension of the SPC activities
is hosted in the new Centre for Structural Systems Biology (CSSB).The 
Scientific Officer – Biophysical Characterization will support users 
with the design,

execution and data analysis of biophysical experiments.

The facility currently covers a broad range of biophysical techniques, 
including calorimetry, mass spectroscopy, circular dichroism, 
fluorescence, microscale
thermophoresis, dynamic/static light scattering, infrared spectroscopy, 
surface plasmon resonance and biolayer Interferometry. The postholder 
will be responsible for
the maintenance and operation of biophysical instruments as well as for 
training users to operate the equipment independently. He/She will have an
opportunity to develop own relevant research activities within the 
framework of the group by Maria Garcia Alai related to the development 
of new methods at the SPC.

I would like to draw attention to the availability of enhanced support 
for sample preparation and characterization for SAXS and macromolecular 
crystallography at EMBL Hamburg. For SAXS, this includes biophysical 
characterization and optimization of samples, access to the P12 BioSAXS 
beamline and, if needed, assistance in data analysis. For MX,
the access includes high-throughput crystallization, automatic crystal 
harvesting and data collection on the EMBL beamlines P13 and P14.

The Hamburg Unit of the European Molecular Biology Laboratory is 
organizing a BioStruct-X Industrial Workshop,to take place
on the DESY Synchrotron Campus in Hamburg from June 15-17 2015. 
BioStruct-X (www.biostruct-x.eu http://www.biostruct-x.eu)is an 
integrated project
funded by the FP7 of the European Commission to support access for key 
methods in structural biology utilizing synchrotron radiation.


The workshop will start with two sessions presenting the facilities 
offered by BioStruct-X partners for protein
production/crystallization and the BioStruct-X synchrotron beamlines 
offering access to industrial users.


In the following sessions, industrial possibilities of macromolecular 
crystallography, small-angle X-ray scattering and medical imaging
will be presented.Here,four dedicated sessions are planned combining 
presentations from the BioStruct-X partners and from the industrial
users of the facilities. A special session will be devoted to the 
presentations by service providers to industry including SMEs and 
spin-off companies.


The workshop will finish with a round table discussion to define the 
future needs of the companies in the use of synchrotron radiation.


There will be 2-3 invited speakers and 2-3 slots for contributing talks 
per session.


The Workshop will take place on the premises of the DESY synchrotron 
facility in Hamburg which operates the storage ring Petra-3, currently
the most brilliant synchrotron radiation source worldwide. A tour to the 
biological beamlines in operation at Petra-3 will be included in
the program. There will be a poster session and ample opportunities for 
networking and advertizing.


The number of participants of the workshop will be limited to 130, and

The main objective of the Course is to teach the young PhD students and 
postdocs from all areas of biology the methods applicable to study 
biological macromolecules in solution. We aim at a comprehensive 
coverage of the field including the major structural and biophysical

structure and structural transitions, macromolecular complex formation, 
protein folding and stability, protein-protein and protein-ligand 
interactions and enzymatic mechanisms. The Course will include lectures 
on small-angle X-ray and neutron scattering (SAXS/SANS), nuclear 
magnetic resonance (NMR), static and dynamic light scattering (SLS/DLS), 
analytical ultracentrifugation (AUC), differential and isothermal 
calorimetry (DSC/ITC) and spectroscopic approaches. Bioinformatic tools 
to analyze protein-protein interactions will be considered, and the 
joint use of the solution characterization methods with the major 
non-solution structural techniques including macromolecular 
crystallography (MX) and electron microscopy (EM) will be covered. 
Special attention will be paid to interdisciplinary approaches, where 
the synergistic use of complementary techniques leads to a comprehensive 
description of macromolecular systems.


The course will be held at the Institute of Biological Chemistry, 
Academia Sinica, in Taipei.


A maximum of 50 participants will be selected to attend the course. The 
registration fee of 40 Euro (NT$ 1600) will be requested from the 
academic participants covering the event materials, lodging and local 
meals. Applicants from industry are expected to pay a 1000 Euro (NT$ 
4) fee.


The Course is oriented towards applicants active in structural biology,
mostly late Ph.D. students and early post-docs but more senior 
scientists, depending on circumstances, could also participate. Students

we are very pleased to  announce the launching of the Small Angle 
Scattering Biological Data Bank (SASBDB, www.sasbdb.org).
SASBDB was developed in accordance with the plans of the wwPDB SAS task 
force to establish a federated system of

interconnected databases for X-ray and neutron scattering (SAXS/SANS).

SASBDB is a curated repository that makes experimental SAS data and 
derived models of biological macromolecules
discoverable, citable and downloadable. Within SASBDB, it is possible to 
browse the scattering data and models,
search by various keywords including macromolecule name, buffer 
composition, experiment setup and author/affiliation
details. 3D representations of low resolution ab initio bead models and 
of the hybrid rigid body models

are available. All data sets and models can be freely downloaded.

Presently, SASBDB comprises of 96 experimental data sets and 151 models. 
Most of the entries contain published data from the studies
where SAS was used for the structural analysis of macromolecular 
solutions (proteins, nucleic acids and complexes). There
are also several benchmark data from a set of well-characterized 
commercially available proteins. The SAXS (and, for some
proteins, also wide-angle scattering, WAXS) data from these samples were 
collected using an on-line purification system to ensure
sample monodispersity. The benchmark set data and models can be used to 
test computational approaches, for tutorials etc.


Researchers are welcome to deposit the SAXS/SANS data and models into 
SASBDB (prior to or after the publication).
The submission requires a simple sign-in procedure followed by filling 
in anon-line deposition form and uploading the data/models.

ATSAS and ATSAS-online users may directly use their credentials to sign in.
The deposition is pre-moderated, i.e. the entries will be validated 
before making them public.

There will be no data collection from own samples at the EMBL SAXS

hands-on experience

characterization
(e.g. using light scattering, size exclusion chromatography and 
thermoflour experiments)

to be analysed

*The course will be held at the Adma Jafet Auditorium at Institute of Physics

University of Sao Paulo (IFUSP), Brazil.*

covering the event materials, lodging and local meals.
Applicants from industry are expected to pay a 1000 Euro fee.

The Course is oriented towards applicants active in Structural Biology,

As you may already be aware, due to unforeseen circumstances related to 
the civil construction of the PETRA III extension, DESY has postponed 
the PETRA III shutdown and user operation will continue until the end of 
2013. Please note that the shutdown is now planned to last for most of 
2014.


In view of this, we announce a call for synchrotron beam time 
applications in biological small-angle X-ray scattering (SAXS) and 
macromolecular crystallography (MX) for the period September - December 
2013.


In biological SAXS, we offer access to an undulator beamline P12 
(responsible scientist Dmitri Svergun). Part of the beamtime will be 
available for soft condensed matter applications through a Collaborative 
Research Group model in partnership with the Helmholtz Zentrum Geesthacht.


In MX, access to the two undulator beamlines P13 and P14 is offered 
(responsible scientist Thomas Schneider).

There will be no formal deadline for submission of proposals, and an 
external Project Evaluation Committee will assess the rolling proposals

We are pleased to announce the upcoming EMBO practical course on
Computational Structural Biology - from data to structure to function

EMBL Hamburg, Germany
15th - 19th April 2013

The course covers computational aspects of protein structure
determination, validation and analysis, including background in X-ray
crystallography, Nuclear Magnetic Resonance Spectroscopy, Electron
Microscopy and Small-angle X-ray scattering and hands-on experience in
model building from X-ray diffraction data, comparing and integrating
different types of structural data, and the differences in
interpretation. Students will also learn to critically examine and
validate data from these techniques.

The course is aimed at PhD students and post-docs working on the
collection and analysis of protein structure data. The goal is to
provide them with insight into the protein structure determination
process, how to critically assess the quality of data from models, and
to provide expertise in the integration and visualisation of data from
different techniques, thus allowing the analysis of protein structure
data for functional relationships.

the EC-funded consortium SystemTB (www.systemtb.org), which aims to 
understand the pathology of /Mycobacterium tuberculosis/ during 
infection. While the core expertise of our group is in structural 
biology, we have established methods in mycobacterial genetics for the 
creation of full deletion mutants and specific point mutants using 
non-pathogenic model systems (M. smegmatis, BCG) (for examples, see: 
Noens et al. (2011) PMID: 2143903, Poulsen et al. (2010) Mol. Syst. 
Biol. PMID: 21439037). We have initialized several new collaborations 
using different --omics techniques, namely in interactomics (using 
high throughput mass spectrometry) (IBB Warsaw), metabolomics (ETH 
Zurich), proteomics (ETH Zurich), and lipidomics (CNRS Toulouse). The 
fellow is expected to carry out and to coordinate experiments in 
mycobacterial genetics in the context of ongoing and future projects.

Electron Microscopy (EM) offered at EMBL Hamburg and EMBL Heidelberg. 
Combined use of these structural techniques will be demonstrated and 
discussed.


The course is aimed at PhD students and postdocs who have at least a 
basic understanding of one (or more) of the techniques listed above, and 
are interested in learning more about hybrid structural biology approaches.

The European Molecular Biology Laboratory (EMBL) is one of the highest

EMBL is seeking an EU Project Manager for the European research 
infrastructure project, Transnational access and enhancement of 
integrated Biological Structure determination at synchrotron X-ray 
facilities (BioStruct-X).


The project will be coordinated by EMBL Hamburg and will include a total 
of 19 partners from 10 European countries and Israel. It will provide 
supported access to state-of-the-art structural biology facilities in 
protein crystallography, small angle X-ray scattering, X-ray imaging and 
protein production. It will be complemented by a series of focused joint 
research and networking activities in relevant areas. The project is 
anticipated to start in the autumn 2011 and will run for four years.


The successful candidate will work closely with the scientific 
coordinator Matthias Wilmanns, the executive committee, all members of 
the consortium, and the funding organization (European Commission). The 
first tasks will be the establishment of appropriate management, data

The Meijers group at the EMBL Unit in Hamburg, Germany is looking for an 
outstanding post-doctoral scientist to work on structural aspects of the 
assembly and organization of the immunological synapse. We use a range 
of methodologies including cell biology, protein biochemistry, protein 
engineering, X-ray crystallography and bioinformatics to study the weak

laboratory has access to state of the art protein production and 
characterization facilities. It will form an integral part of the new

Job description: The primary duty is to implement the scientific

Qualifications and experience: The ideal candidate will hold a PhD in 
natural sciences and will have minimally two years of postdoctoral 
experience in structural biology or a related research area. Previous 
know-how in research training and/or the development of training 
materials will be an important asset. Experience in designing and 
running scientific courses from introductory to advanced levels is 
necessary.


In addition to appropriate project management skills, the successful 
candidate will have excellent organisational and communication skills as 
well as the ability to contribute effectively to a team. Familiarity 
with Windows, MS Office, e-mail and Internet applications is essential. 
A working knowledge of other operating systems (e.g. UNIX/Linux) is 
highly desirable. HTML editing and designing, as well as knowledge of 
Adobe Creative Suite, QuarkXpress or other drawing and page-layout 
programmes are considered to be an advantage. Fluency in English is 
essential and familiarity with other European languages is desirable.

PH.D. AND POSTDOCTORAL FELLOWSHIPS ON STRUCTURAL/FUNCTIONAL 
CHARACTERISATION OF PROTEIN INTERACTION NETWORKS IN YEAST


Two Ph.D. fellowships and one postdoctoral fellowship are available in 
the research group of Matthias Wilmanns at EMBL-Hamburg, Hamburg, Germany.


Src Homology 3 (SH3) domains are modular protein domains which are found 
in hundreds of multifunctional proteins from the human genome and tens 
of similar proteins in eukaryotic model organisms such as S. cerevisiae. 
Many of these proteins are involved in signalling and the organisation 
of the cytoskeleton, for instance. Although the structure and canonical 
binding site for proline-rich ligands has been well studied over more 
than a decade it remains largely unknown by what kind of mechanism many 
of known SH3-domains containing proteins are sorted to correct protein 
ligands within complex cellular networks, to allow their proper function 
in living systems.


The key aim of our future research will be to identify the molecular 
structural basis of cellular protein networks that are mediated by SH3 
domain containing proteins, using four yeast species (S. cerevisiae, S. 
pombe, A. gossypii, C. albicans) as models:


1) We are planning to determine structures of SH3 domain containing 
multifunctional proteins, to unravel their domain organisation, which 
may be important for correct folding and regulation of SH3 domain 
mediated protein/protein interactions.


2) We are aiming to determine structures of SH3 domain mediated 
protein/protein complexes, to provide insight into the molecular 
organisation of SH3 domains within complex protein networks in living 
systems.


3) We will exploit our structural data to explore the use of SH3 domains 
for drug discovery approaches, using both computational and experimental 
screening methods (a large depository of structures from S. cerevisiae 
is already available).


4) We will probe structure-based function by in vitro and in vivo 
approaches, by employing genetics, imaging techniques, and cell biology 
approaches.
