Hope following information helps you:
1) I am going to use the anti-virus in a closed network, and connection to
the Internet is not possible. How can ClamAV be updated manully without
accessing the Internet? Can "freshclam" be deactivated? And will there be
any effect on the signature update if "freshclam" is deactivated?

==> You can download the Virus Definition manaully from the internet and put into your server and then reload it, the new virus defintion will reloaded into ClamAV. And surely, freshclam can be deactivated manaully by stopping the freshclam deamon.

2) There is a large number of end users within the closed network. Are mass
installation and update possible? Are remote installation and update
possible? If yes, how can they be done?
==> In fact, you can setup a update server for such Virus Definition Updates.


3) What is the actual virus database update frequency?

==> I am not so sure, it seems it's daily. Or even hourly, ClamAV repsonse to new Virus very fast. In my case, i set the server to update 2 hours a time.
