What isn't clear is if I need this if I have auto updates turned on?

I had Mac OS X, 10.5.3 on one of my computers. I downloaded a combo
update that contained all of the updates from 10.5.4 to 10.5.7 and
installed them in one package.
Does Microsoft offer combo updates for Windows?
Depends on who you ask. Sometimes the SP's will rule up previous
updates in their package.
I usually prefer to do them one at a time to make sure it works.
But read the package literature first.

Typically service packs have been rollups of all previous SPs for that
OS and didn't require anything beyond RTM. I

You must have Windows Vista with SP1 or Windows Server 2008 installed
on your computer before installing Service Pack 2. (Windows Server
2008 shipped with Service Pack 1 included.)
