I just want to say that this hit my Stretch setup by surprise today after
rebooting also. Basically this is the default setup with dnsmasq so the
package is basically broken in Stretch at the moment. Any chance that this
will be fixed before Stretch goes stable?

I just want to say that I got to troubleshoot this issue in a saned
setup that broke after dist-upgrade to Jessie. I ended up renaming the
service file to a template-definition (saned@.service) since that was
indicated in the error message, but I didn't know about the
single-instance stuff.
There is some movement on this bug other places. It is now fixed in Ubuntu:
