Thank you for opening this thread, I personally deployed and successfully served an App Engine Flex custom runtime application following this documentation <https://cloud.google.com/appengine/docs/flexible/custom-runtimes/quickstart> to the southamerica-east1 region. I would assume that the issue may be in the way your application is handling requests that being said, the GCP Google Groups are more oriented towards general opinions, trends, and issues of general nature touching App Engine. I would recommend bringing this issue to a website such as Stack Overflow <https://stackoverflow.com/questions/tagged/google-app-engine>, where experienced programmers are within reach and ready to help.
