We cut a decent number of unstained as well.  Mostly the tiny biopsies and 
cores and we do end up using most of these. I personally see no reason to cut 
unstained slides on anything else.  Since you still have to go back and cut 
controls, it doesn't really save that much time but of course, it does save 
tissue.

I have had experience in three different labs.  The unstained slides can
pile up quickly, especially when you are using charged slides that are
just in case slides for sendouts, IHC, etc.  For core biopsies (breasts,
fine needles, prostate, etc.) we cut levels and put up at least five of the
in between sections to keep on hand in case.  We maybe use these slides
10 percent of the time and toss the rest after 3-4 weeks.  It can seem
wasteful, however if you have a small core, you must save precious tissue.

[Histonet] mouse brain cryosectioning help

Hi Histonetters, I need your expert advise on how to get good  mouse Brain 
cryosections. 
We are getting folds, cracks and wrinkles. It may be due to mushy nature of 
Brain tissue. Any suggestions to eliminate those artifacts.

I do the same thing.  Adding a couple of drops of paraffin over the label has 
worked for us for years.

If you keep the layer of paraffin thin enough, it can still be scanned with 
most barcode scanners.

Just in the process of starting a new AP/histology lab myself, and what I used 
as my main resources to develop the quality metrics, QC and QA were:

 *the CAP/ASCO guidelines,
 *CAP checklists, HistoQuip studies,
 *   Quality Management in Anatomic Pathology, ASCP publication, 2003 ( forget 
the exact title),
 *CLSI publications QA for Design Control and Implementation of IHC 
Assays, 2nd edition,
 *   CLSI Fluorescence In Situ Hybridization, methods for Medical Genetics, 
Approved Guideline.


 United the standards in the SOP's and LIS documentation/reporting. Overall, I 
tied them to measures of timeliness, quality of histologic preparations, test 
phases and benchmarks, competency.
 Monthly quality meetings, occurrence management with process control ( RCA, 
etc), Pathologist review documentation.

[Histonet] Tissue Processor

We always validated our processor but not formally.  CAP wants a more formal 

approach with procedures. 



What we did in the past was run placenta and colon and reviewed stained slides. 

We may have to be a little more critical and run various types of slides and 
programs.
