[Histonet] Sucrose cryoprotection

2018-04-05 Thread Mike Tighe via Histonet
I'm having problems getting good cryosections of mouse lung tissue that has
been formaldehyde fixed, cryoprotected with sucrose, and embedded in OCT. The
lungs were fixed with 3% formaldehyde for 24 hours followed by 10%, 20%, and
30% sucrose until the tissue sank in solution. When sectioning the tissues
collapse. I have tried cutting colder and cutting thicker but still getting
poor sections. I embedded whole lobes of the lung individually.

Any thoughts would be greatly appreciated !!

[Histonet] RCC control blocks

2018-04-05 Thread Charles Riley via Histonet
Does anyone have any RCC control tissue they are willing and able to share.
I only have one tissue sample and it is showing a lot of non specific
staining. I think it wasn't properly fixed and the pathologists don't like
the results.

We do not run the test very often so management doesn't want me to waste
money on buying control slides. One small piece of tissue could last us a
long time and hopefully we get something in the interim.

Any help would be greatly appreciated. Thank you in advance
