I am currently running a research project and the researcher wants me to use 
Caspase-3 for apoptosis staining. I have never worked with this antibody before 
and want to know if anyone has any suggestions on what to use for a positive 
tissue control and a negative tissue control?


I purchased the antibody from Novus Biologicals and they recommend a Jurkat 
cell lysate for positive control but I am not familiar with this as well.

Any guidance would be greatly appreciated

[Histonet] Question for Research on Bone

So I am working with Canine femurs and tibias that have been dissected as 
medial and lateral - they have been decaled in formic acid - rinsed in running 
tap well - processed on a routine 1 hour processing program . cutting very 
well!!

My issue is that the articular cartilage on the bone surface keep flipping up 
and over onto itself - I am using a 37C slide warmer overnight to allow them to 
hopefully flatten out more than on the water bath and then drying in the 56C 
oven for about 1 hour - then staining on automated Leica stainer - they are 
intact at 37. look like the might be curling at 56C??

Anyone have any suggestions???

[Histonet] Research IMF help

To all - I am working with NBF fixed paraffin embedded tissue and need to do 
IMF staining potentially using two different pH retrievals, that are all the 
same primary host species - I know that if I am doing bright field, I would 
retrieve, stain for primary 1 with DAB first and the sequentially retrieve and 
stain for primary 2 with a different chromogen.. any suggestions when 
working with fluorescence?  Does the first reaction get removed from the 
retrieval for the second epitope; I am in new territory and could use some 
recommendations. TIA.  Amy

You need to add sucrose to your PFA we use 4%PFA+4%Sucrose to fix and then 
cryoprotect in 30% sucrose which all need to be prepared in Phosphate buffer 
solution NO saline  if you message me directly I would be happy to share 
our SOP this was a very hard thing to learn not a lot in technical 
references about this process.

Fix in 10% NBF - 10 minutes
Rinse in running tap water - 5 minute
Rinse in distilled water
Gill 2 Hematoxylin - 1 1/2 minutes
1% Glacial Acetic water - 3 dips
Running water - 2 minutes
(we do not need bluing agent - our water takes care of it)
95% Ethanol -2 minutes
1% Eosin/Phloxine - 2 minutes
95% Ethanol - 2 minutes
Remaining dehydration in absolute ethanol & xylene - 2 mintues rest of the way 
down

I don't see a problem with a research setting. If it was patient care, CLIA 
would need to inspect the set up in the person home. The same goes for our
