Clicking on the window first does in fact make it possible to use Ctrl-O to
open a file. But of course I might just as well click on "Open File" if I have
to use the mouse and click on the Window first.

You're correct. It does work correctly in safe mode. It's strange, though,
because I have the same behavior on two separate computers, one an ASUS, the
other a Toshiba, both running Windows 10. The only additions I made to the
default configuration was a Portuguese spell dictionary and Portuguese
interface (though I use English most of the time), plus I removed the French
and Spanish dictionaries/interfaces and turned off auto update and fast start.

Perhaps a Windows 10 only issue?
Did you try to run Libreoffice in Safe Mode? (Help > Restart in Safe Mode...)
