The problem is that FontHook has to redefine the font synonym
'LettrineFont'. You have to do it like this:

  \unexpanded\def\myfonthook
{\definefontsynonym[LettrineFont][SerifBold]}

  \setuplettrine[Lines=3,TextFont=\it,
FontHook=\myfonthook,
Findent=0pt,Nindent=0pt]
