I assume you meant to say rc.local, as this has nothing to do with iptables nor the qubes-firewall-user-script. In any case, this suggestion in itself is a huge hammering. The sys-firewall shouldn't have to change the qubes-uplink-eth0 (Network Manager configuration) in the first place and that
