IMHO it isn't a vpopmail problem. It is an message from qmail-smtpd.
Check the patch applied to qmail-smtpd. Probably you patched the qmail and you don'tcheck the README before.
Another question, Do you know if the sender domain is valid?

I have a fresh install of qmail/vpopmail. For about 3 months now, I
have had users logging in a getting/sending mail with no problem. I have
gone over LWQ many times and I don't know why but when sending email to
some users I get this in the log:

There's only one file in users/. The file is .vpasswd.lock.
