Hashtag is the actual and correct term. it's us screen readers who have been hearing it wrong the whole time. It could even be considered an accessibility improvement/bug fix. In the PC Chicken Nugget Twitter client, I even included a Jaws dictionary manager entry to have the number/pound sign pronounced as hashtag. It's most likely one of those things you'll just have to get used to. If you're dead set on having the number sign misspoken as number in that app, then I don't think there is a means of accomplishing it.
