The way I understand it - and I may be wrong - would be something like: <dfn title="This is the definition">term</dfn> Semantically, a bit like an inline, single item definition list. In its application, similar to acronym or abbr, to expand the meaning of a term (as opposed to an acronym or abbreviation, of course). Topically enough, this was just mentioned over here http://www.accessifyforum.com/viewtopic.php?t=2111 So, you'd have it when you have complex terminology that you'd like to expand upon inline (although I'd much rather have something like a separate glossary section, as I'm not convinced about support for dfn in things like screenreaders - and even visual browsers, when you're not hovering over a term with the mouse, don't normally expose definition title attributes).
