Of course you can ;-)
You need a standalone installation of Jython and then it is just the
same:
jython myscript.py
Look into the Jython docs for the environment variables to be used for
addings to class path and sys.path.
Another possibility (no need for an extra Jython installation):
put your myscript.py into a folder myscript.sikuli and use
java -jar sikuli-script.jar myscript.sikuli
the paths have to be adjusted to your situation.
