The Cisco method is documented here:

I think your "Signal= '2'" should be changed to "Signal= 2".

Also, this may be obvious, make sure you have an empty CRLF line between
the SIP message header and message body.
So, showing invisible characters, you should have:
Content-Type: application/dtmf-relay<CR><LF>
ContentLength: 26<CR><LF>
<CR><LF>
Signal= 2<CR><LF>
Duration= 250<CR><LF>

Note also that the content-length above is 26.
