I should think so.  I use evolution and mutt together on the same
mbox mailfiles.  evolution from my desktop machine at home using
imap to the mail server (keeping all mail on the server) then when
I go into work I "empty trash" and exit evolution then from work
I ssh in and run mutt directly on the mailserver.

If I forget to exit evolution then when I get back home it will
complain about corrupted mailfiles cos mutt has changed them
underneath it.  So long as you only use one mail reader at a
time it should be ok.
