I assume you are going to support the plexi-glas on some kind of
framework.  You can use double sided tape to hold the plexy to the wood
supports, and fill any gaps in the plexi-to-plexi or plexi-to-pattern
joints with modeling clay.  Roll up a 'snake' of clay and press it along
the joints with your fingers, try to get it into any gaps.  Use a single
edged razor blade to strike it off flush on flat surfaces, and use the
corner to create a sharp corner in the parting surface-to-pattern joint.

Why use plexy?  Soft pine or other smooth, long grain clear (no knots)
wood works very well and is easily fitted to the pattern,  and can be
finished with lacquer sealer in a short time.  Any imperfactions on the
original pareting surface will be matched on the second surface as it will
be cast against the first one.
