4) Does user filter have access to /usr/local/sbin/spamfilter?

Do me a favor - not that this should really matter, but try changing:
smtp      inet  n       -       n       -       -       smtpd -o 
        content_filter=spamfilter:

To:
smtp      inet  n       -       n       -       -       smtpd
        -o content_filter=spamfilter:

Don't forget to reload Postfix.
