Usually autocomplete fires up automatically when a . or ( is typed. Looking 
at the code I can't immediately say how a completion is implemented (other 
than "it looks like it is provided by rope"), so I guess somebody should 
describe that and/or implement debug hooks that print what's going on with 
completion when Ctrl-Space is clicked and doesn't work.
