After some investigation, I found the culprit: Avast (which I only have installed in this particular machine).
The anti-virus company was recently bought by Norton. As expected, there were changes - so the company released "a new version" of their software a couple of weeks ago. Their malware shield was - in a "stealth mode" (i.e., differently from Windows 10 native antivirus, it didn't provide an alert about the action) - quarantining some .exe files. I might reinstall Avast again in the future (but will have to find out how to allow specific files to run). But as of now, I uninstalled it - the Spyder update (in a new environment) worked.
