Go into Settings -> Basic Settings tab. What does it show in the "Media Folders" for your music folder? Sometimes when a drive is removed and then re-plugged it may get a different drive letter than it had previously. Verify that what you have specified here matches what Windows Explorer shows for your drive/folder where your music lives.
