Yes, it does.

Use an external_acl_type and set the TTL low.  Doing so is going to have a 
negative effect on your performance.

Running a process in the background that monitors the acl file and runs "squid 
-k reconfigure" when said file changes would likely work as an alternative.  
Other tools may be available to perform the service you are looking for with 
more grace.
