Try:

MAILDIRFILTER=./mailfilter
MAILDIR=.

And you'll have to add a .qmail file for your user that passes all
of your mail from vdelivermail off to maildrop, otherwise, nothing
will get filtered.

Read up on 'man dot-qmail' and read all of vpopmail's documentation
before attempting to create the .qmail file for maildrop. This is
basic required knowledge.

A system-wide maildrop implementation will require using maildrop
as your primary delivery agent. If you want to use maildrop with
vpopmail, then you're going to have to create a shell script that
retrieves each vpopmail user's homedir from vuserinfo, and then calls
maildrop appropriately to deliver the mail. 'man maildrop' and 
'man dot-qmail' and 'man qmail-local' will be useful here.
