Hello,
1) I assume floor size refers to the area that is covered by the vehicle
trace. If the map is large, a few vehicles may not cover the whole bounding
box. Thus, the bounding box may change when there are more vehicles. I
would expect the bounding box to strictly increase with more vehicles and
find the change from minx 3915 to minx 4181 a bit surprising. I suggest you
use traceExporter.py with the option --poi-output and compare both traces
visually in sumo-gui.

2) when using option randomTrips.py with option --route-file, duarouter is
called automatically. (you may call it manually on the trips)
If you load the trips directly into sumo, you will probably get a better
distribution of routes (vehicles react to congestion).
If you use duarouter you should consider
http://sumo.dlr.de/wiki/Demand/Dynamic_User_Assignment

3) we have some small-scale scenarios based on real-world data at
https://sourceforge.net/projects/sumo/files/traffic_data/scenarios/Bologna_small/

regards,
Jakob
