If your two machines are the same OS version, and have the same drive structure, yes. I do it all the time, just copy the entire contents of the profile folder.
If the OS versions are different (say Win 7 to Win XP) you can still do this but you'll have to edit the pref.js file on the target computer to adjust some of the paths that will be different.
