That's generally the purpose of the "omit" box.  At the stage of the search
at which you want to issue the command to exclude people named james, enter
james in the name field and click 'omit' and this will give you everyone
else specified by other parts of your query...except the james people.  

Keep the 'omit' requests at the tail end of your queries.
