With Debian Jessie's libvirt + virt-manager + KVM, one can emulate a USB device and flag it as removable, so that Tails accepts to boot from it (and to create a persistent volume on it). That's documented there: https://tails.boum.org/doc/advanced_topics/virtualization/virt-manager/index.en.html#index5h1 (I didn't test these instructions myself, but I trust sajolida to have done it himself.)
