We're not running 2010 but I know that in our case if I have a script machine (or any pc for that matter) that is going to utilize SMTP to send mail outside of our domain we have to identify it within exchange. Not sure if that was in 2000 or not but I know it's something I have to do with my machines in order to email some outside vendors.

Sounds like a problem with your SMTP settings. If your e-mail server was upgraded from Exchange 2000 to Exchange 2010, perhaps the server name has changed or now it's using secure SMTP.
