Two possibilities to check: First, be absolutely sure that you don't have a copy of MixW already running. It is very easy to try to start a second copy by accident.
Second, go to (in Windows) Control Panel > System > Hardware > Device Manager and look under Ports (COM & LPT) to see for sure what the number of the USB Serial Port you are using should be.
