My preliminary testing suggests that it doesn't support variables or macros. At least the simple tests I tried wouldn't work. So if you want a TOC, for instance, it probably won't work. It does appear to support [[*]] links, so that may be more important to some people at the moment ... but down the road not supporting macros may be a problem if you want to use the automation features of TW. Is anyone else having troubles with the tw5-markdown plugin?
