Hello I had exactly the same problem when I upgraded to Opera 10.50.
I managed to get it working by copying the tiddlysaver.jar file into a directory that was already in my Java classpath. I think Opera has just switched to using the Java plugin and it couldn't find tiddlysaver.jar on my desktop as the Desktop directory was not in the classpath. I hope this helps.
