Simply create a space where you want to import it, go into the backstage click on the space tab on the left and use the import tab. Note that if your layout breaks you may need to fix it manually to work with TiddlySpace. Going into the includes tab and removing system- theme may help with this.
Currently support for going the other way (taking a space offline, working on it and uploading again) is very bad. You can obviously download the space and it should theoretially work (with TiddlySaver.jar for certain browsers) however certain things are likely to break in offline mode. I can assure you that supporting all of this is definitely a TiddlySpace goal. The best way to get things happening is to start using it and demand new features for it based on problems you encounter :). So good luck and let us know how you get on!
