Most of the "programming" I've made with TiddlyWiki is moving around with buttons <<newTiddler>> <<search>> etc . Then I've made heavy use of the tiddler macro for embedding one tiddlers content into another - like this: In tiddler [[B]] I write <<tiddler A>> and tiddler A is rendered into tiddler B. This tiddler macro is *very* usefull because you can use it as a template. I.e Tiddler [[A]] says "This is tiddler $1" and in tiddler [[B]] I use the tiddlermacro like this: <<tiddler A with: B>> and it renders "This is tiddler B". Simple yet very effective - also for very intricate templates... There are a lot of wonderfull plugins for TiddlyWiki which extends it's capabilities, and if you read through the documentation, you'll get an idea of how to use them. Checkout TiddlyWiki googlegroup's threads to find plugins you'll need - jump to http://community.tiddlyspace.com/ to get some pointers...
