Maybe I can help with two of your points:
2) I already have a list of wikis on the "community search" wiki at
https://rawgit.com/erwanm/tw-aggregator/master/tw-community-search.html#CommunityWikis.
It is limited to the wikis for which I have the author's agreement to
extract their content, but I hope to expand this list ;)
4) this is very similar to what my "aggregator" scripts do, it would
require only a little adaptation. I don't know if it is possible to
automatically push the tiddlers into a tiddlyspot wiki, though.

Either way it would be awesome if the authors could use pull the
tiddlers used to make and edit the cards from my site and create their
own cards that I can import into the list.
Here is the listing, the tiddlers needed to make the cards and a
filter to use for the nice batch export feature in the advanced search
are listed under the 'Tiddlers Used for TWcards' tab.
If I am able to convince people to actually use this than the next
steps are:
1) Make a dedicated wiki on tiddlyspot for a general directory
2) Make a tool to also list existing wikis as a wiki directory
3) Possibly change the capitalization to twCard instead of TWcard in
the things I have made because that seems to be what other people are
using
4) Figure out if it is possible to make a script that, given the urls,
will check a specific tiddler or tiddlers from a set of wikis and
import the changed tiddlers so that plugin authors just need to update
their local copy of the twCard and it will get pulled to the directory
by a script
