Having a good understanding of the basics of HTML and JavaScript*
(including the DOM) is the first step. Some knowledge of CSS might also
come in handy, though that's not necessarily required.
Getting familiar with the framework - i.e. TiddlyWiki and/or jQuery -
can be a gradual process.
It helps having a specific need/task and gathering the required
knowledge in piecemeal fashion. (To this day, I don't have a perfect
understanding of every single component of the TiddlyWiki core.)
Looking at other people's code (e.g. plugins) is an important part of
this process.

* Note that Java is very much different from JavaScript, and barely
relevant in the TiddlyWiki universe; only the TiddlySaver applet is
written in Java.
