Here are some large installations I'm aware of, thought I'm probably
forgetting some:

* Actrix Networks, the largest ISP in New Zealand has provided some
preliminary statistics[1] on their TMDA usage.

* mail.gnu.org, which handles the Free Software Foundation's mailing
lists sends a few thousand[2] messages per day though TMDA.

* Gmane runs a TMDA service[3] which sees lots of use periodically.
For example, it handled 6GB worth of messages this past weekend.

There are some other TMDA providers listed at
http://tmda.net/inuse.html, though I don't know how many of those
qualify has ``large-scale''. That list also isn't comprehensive since
most sites don't bother with being listed there.

I don't see resource consumption being a problem for a few thousand
users, espeically given how cheap hardware is these days. Allot
depends on how you've tuned your TMDA configuration. There are things
I definately would and wouldn't use were I desgining a large TMDA
installation. Also the defenses you are using in front of TMDA will
make a big difference. e.g, virus scanning and reasonable SMTP-level
spam reduction will make TMDA's life much easier. Even using a
content filter like spamassasin might be something to consider.
