You are missing the "x" on the directory assuming that the access is
occurring as "vpopmail."  On a directory execute allows access.  Right now
you are denying access to the filters directory.

Just to be safe would you mind changing the permissions on your filters
directory to 755:

% cd /home/vpopmail/domains/nesys.it/test/.tmda
% chmod 755 filters

If you had done the "cd filters" as a user other than root, you should
have received a permission denied.  If vpopmail has a valid shell you can
test access to files by su'ing to vpopmail and checking if it can read
them.
