If I understand correctly, the incoming filters are searched for first
match from top to bottom. So if there is a blank subject line then the
filter you have created will already work. But if there is no subject
line, you might need an additional filter after that one to catch it.
Is there a way to use a NOT expression, as if to say that if there is
NO header that starts with "Subject:" then to bounce it?

Not sure about syntax, but something like:
headers (Subject:.$) confirm=blank_bounce.txt
headers !(Subject:.$) confirm=nosub_bounce.txt

Not sure if that would work, but just an idea. Sure would be nice to
hear from the TMDA creator on this one. Has ANYONE heard from Jason? Is
this the last TMDA version that will exist?
