First off, one post of a question is enough. Second, your question is really not entirely clear. Are you asking how spamassassin is called via qmail-smtpd? If so, the QMAILSCAN environment variable is set in ~vpopmail/etc/tcp.smtp, which tells qmail-smtpd to call simscan. Simscan is the scanning harness, which calls clamd and spamassassin, and also does some of its own content scanning (attachment blocking, optional regex). So if you followed the toaster install, you don't have to "tell" spamassassin to look for emails. It should already be doing this, and you can verify it by looking at /var/log/spamd/current.
