A very good FREE anti-virus product is Microsoft Security Essentials:

One option they offer is to download a boot thumb-drive or boot CD image with the anti-virus program built in.

Booting from something other than the system drive is particularly useful to remove "Root Kits".

When your PC is set to boot from removable / external drive ( USB Thumb Drive or CD/DVD ), and you insert the USB Thumb Drive or CD/DVD and reboot, the screen will be blank for a few minutes, the a monochrome Windows logo will be displayed, after a while you will get a report of what malicious software, if any, was found and what was done to remove it.
