what kind of file, is it by any chance ALAC or AAC ? If i would hook an USB drive to the Touch i would use flac files .
ALAC/AAC is a bit buggy at the moment (see the dozen treads already open) . And also I -think- native ALAC playback on the Touch is not enabled at the moment, because of some issues.
