Your activation email may have been discarded because it had a wrong From line. And, yes, it requires a configuration in pootle.prefs. (If you did not receive the mail, you can also find the activation code in pootle.prefs or users.prefs) I recommend you to read /usr/share/doc/pootle/README.Debian for the detailed steps of the setup of a Pootle server on Ubuntu.
