Normally most Flash devices I know (SD Card, Micro SD Card, USB Stick, CF Card in Reader) will behave like a usual HDD. Of course you have much lower writing speed but when using the Trisquel you will not notice anything. I run Trisquel 5.5 from a 16GB USB stick without issues.

Only modifications compared to a HDD installation so far:
- Noatime option for mounted filesystems
- No swap partition
- Moved some directories to tempfs

I can not comment on the average lifespan of a USB stick in daily usage but so far I haven't experienced any issues.

Your hardware must support booting from USB devices of course but this is what most modern hardware does.
