Anyway, your error was to omit a space between 777 and /home/john/virtualbox and to write /Home instead of /home. But, again, there is no notion of permission on FAT filesystems. That probably explain why you cannot change them.
Are VMDK files supposed to be directly executed? I doubt so. Besides, VMDK
files are for VMWare, which is proprietary software. You can convert them to
VDI files for VirtualBox, which is mainly free software (entirely if you
install it from Trisquel's repository).
