It depends on what you want to do. There is no single package I know of that includes *all* the Python modules available - there are too many and many are out of sync with different versions. If you do scientific/numeric computations you probably want something like Anaconda or Enthought. If you do media/video work you might want some of the distros targetting Maya or similar. Of the "standard" distributions I usually recommend the ActiveState.com distro because it includes some extra Windows goodies and integrates with the help system better. If you need an IDE you will need to check those out separately, there are at least half a dozen, some free, some commercial.
