Unfortunately, no; I cannot provide instructions on how to reproduce it, as I'm not 100% certain how it happened in the first place. I have a suspicion it has something to do with the migration to the SQLite DB backend, but I'm not certain of that.
It happens for me routinely, though. And no, the problem is just a mismatch between the folder view and the main list; usually (but not always) on startup it reports the number of unread messages it first finds accurately, whether it is one or multiple; after that, it fails to update the figure.
