Try right clicking on the volume control applet, and going into the settings window. Then go to the hardware tab, and click the drop down box you see towards the bottom of the window. Make sure that you have a profile selected that includes input of some kind, usually analog output + analog input duplex. Once this has been selected, try using your mic again.
